When building a riser or stage is it necessary to nail or screw the stud supports in place or can you just insert them inside the frame. Mine will sit on top of the existing carpet in the room so is it necessary to fill the space in between the studs with insulation as well. I am prepared to do either/or with both questions I have.

You should fix them to the frame somehow, whether it be screws, nails, or joist hangers.

I would fill the riser would something, having an open cavity sitting in the middle of your room is probably never good ;) Just stuff it with some cheap insulation and be done with it.

I'm not quite sure what you mean, but it doesn't matter: I would not have any pieces of wood in your stage/riser that are not nailed/screwed/glued down somehow.

Secure them to the side framing. They can just sit on top of the carpet.

Yeah, secure them. "L" braces at least are not expensive to keep them in place.

Not sure you need braces, Just use deck screws through the outside frame into the stud built "mini walls".
