My at grade Theater will be on a concrete slab and the Builder put down 6 mil plastic before he poured the slab so I don't see any reason put down more plastic before I carpet the room.
I may put down Dri-Core but am not even sure I need that since the room is grade level and I should not get any dampness.
What do you all think?

Did he also put down rigid insulation board and wire? Hope so. Anyway, I would not put down carpet directly on concrete, its uncomfortable and it not a good practice. What I would do is put down some type of sub floor either by using Dri-Core or conventional sleeper with a subfloor.
In the midwest, I have seen a lot of basements finished where you have a pad and then your carpet right on the concrete. My house will be one. If your basement is dry, it won't be an issue.

Here in VA, it is standard practice to put pad and carpet directly on the concrete slab.

As long as the basement is dry, padding and carpet on concrete is fine.
My house is on a slab. no basement, the Theater is on the first floor was just wondering whether additional plastic would be needed since it was already placed on the ground before the slab was poured.
Here in Atlanta we don't have that many below freezing days.

I am near Atlanta and it is not necessary. The plastic is a moisture barrier and you should not need 2. Just put a GOOD pad down and carpet on top of that and you should be fine.
I put down the thickest foam pad I could buy. Worked great.
