Calibrations is setting up the user accessible and advanced image controls on your set to produce a picture that is as close to the Rec 709 HD Standard that was used when the content was mastered in the studio.
This involves the use of special display patterns, meters and software to adjust the set to match the standards.
Sets are not typically calibrated from the factory on purpose although some of the newer sets that are THX certified come close.

Will you see the difference? Maybe! It depends on how interested in image fidelity you are. Once you watch a properly calibrated set for a week or so, you will be able to spot the non calibrated (almost every other set you will see) in an instant.

What does it cost? To have one of the pros that have high end gear, know what they are doing and can spend the needed time to do it correctly, as well as pass on an educations, $325 - $450+ depending number of inputs, capability of the display etc.
I have paid to have this done, and since invested the time and money to be able to do this myself. Projector owners need to perform a calibration more often since the lamp in a projector changes as it ages. Flat panels change a little but not that much. Most calibrators will offer touch up servers for much less than a full calibration..
