One connection should be sufficient, and I'd use the sub out into the LFE input of the sub if it has one. What sub is it? What AVR?

What i meant is, i have two system , a 2 ch dedicated pre+ power amp and an AV but only one sub, one sub . i want to use the sub for both, how do i connect using one connection.? It would be much easier to connect it from diferent out ( indepedely from the 2 ch and AV sub out, since the sub already have a pair of input rca.
im not sure if that would creat intreference.

In that case since they're both line level outputs shouldn't be an issue but you probably wouldn't want both preamps on at the same time. If it does create an issue then you could get a line level switch. Still depends somewhat on what the specific input types you have on the sub and how you would set up the crossover...
