The sealing aspect should work immediately and cure within a few hours as you are essentially just filling a gap with a flexible material. However, the question you should be asking is which acoustic sealant stays flexible the longest after initial cure without pulling away, cracking or drying. To that extent, Kinetics Noise Control has tested virtually all available sealants in their lab and have determined that Sikaflex 1A outlasts all others in this respect - by far. http://www.amazon.com/Sikaflex-Polyurethane-Performance-Elastomeric-Limestone/dp/B00461GOKY

If you filled big gaps it will take a couple of days for the caulk to cure, It may shrink in the process and another coat would improve the integrity of the seal. This often happens at the bottom of the wall where the drywall sits above the concrete.

Are you having concerns about the level of soundproofing you've acheived?
