People usually use the resolution closest to the tvs native resolution, so in your case 1080i for the cable box and 1080p for the upconverting dvd player would be the preferred settings.

I have a new 40-inch 1080P Samsung LCD.
What are the best settings for Cox HD cable and OPPO up-converted DVD?
1080i or 720P?
The max on cable is 1080i.
The Oppo up-converts to 1080p

Is it really better to go from 1080i to 1080p vs. 720p to 1080p?
That is what I use and I like it. But just try both and see what you like better.
