I want try a series of calibration readings using off the screen measurements as a reference offset, then applying that to readings directly from the projector.

Where should I position the meter when facing the PJ? Next to the screen? Closer to the projector?

I'm using ChromaPure and an i1 Display LT (with diffuser for the off the lens readings).

The only reason to do this is to get higher light output from the lens reading. The closer to the lens the more intense the light. There's no rule about this, other than don't get it so close that it overloads the meter.

BTW, unless you can't get a good reading off of the screen, I wouldn't do this at all.
