What filetypes support TrueHD/DTS-HD?

For some reason I'm having a hard time finding this answer online...can files like mkv hold trueHD/DTS-HD audio if played through an HD4850 over HDMI to my receiver?

Not per se. MKV can hold multichannel FLAC which sounds the same. You need to have a .EVO or .M2TS to hold the encoded audio stream. These are the native file structures of HD-DVD and Blu-Ray
